Role and Importance of Mitophagy in Skeletal Muscle in Health and Disease

Mitochondria are organelles providing a large part of energy to cells via oxidative phosphorylation. They form a dynamic network, the abundance, size, distribution and morphology of which are continuously rearranged through biogenesis, motility, fusion and fission events. In addition, severely depolarized mitochondria are removed through mitophagy, namely a selective form of autophagy. While mitogenesis, fusion and fission events have been quite extensively studied, much less information exists about the specific role of mitophagy. Excessive mitophagy contributes to mitochondria loss and muscle dysfunction in some diseases. On the other hand, it has been hypothesized that moderate mitophagy could induce mitochondrial biogenesis in skeletal muscle but this hypothesis is nowadays still under debate. If the latter reveals true, new therapeutical perspectives will be opened to stimulate mitochondrial biogenesis in this tissue. It is therefore essential to thoroughly evaluate this hypothesis and to see to what extent the role of mitophagy can be a critical signal for mitogenesis and other adaptations in the mitochondria. The aim of the symposium would be to emphasize the role of mitophagy in skeletal muscle in health and disease.
